Først i en alder af 19 år begyndte Nicki Sørensen at cykle, men derefter gik det stærkt.

I 1999 underskrev Nicki sin første professionelle kontrak med Team Chicky World og da det lukkede skiftede han til Team Fakta, hvor han med flotte præstationer fik bevist sit værd.

I 2001 skiftede Nicki til Team CSC – World Online, som Team Saxo Bank hed dengang. Med stærke præstationer kørte Nicki sig på Tour de France holdet og der blev han de følgende fem år.

2003 blev året hvor Nicki for første gang vandt det danske mesterskab og som altid var en del af et meget stærkt Tour de France mandskab.

I 2005 var et meget succesfuldt år for Nicki med sejr i GP d'Ouverture la Marseillaise og en flot fjerdeplads i Tour Méditerranéen. I september vandt Nicki 18 etape af Vuelta a Espana, efter at have spurtbesejret Pascual Rodriguez.

Det danske mesterskab blev også i 2008 vundet af Nicki, men det var året efter, at karrierens største sejr skulle komme. På tolvte etape af verdens største cykelløb, Tour de France, viste Nicki hvem der var den stærkeste i udbrydergruppen, da han hel uimodståeligt kørte væk, og vandt karrierens til dato største sejr.
